  • NEXTflex ChIP-Seq Kit for Improved Genome Wide Analysis of DNA-Protein Interactions

Bioo Scientific announced the launch of the NEXTflex™ ChIP-Seq Kit which was designed to simplify the genome-wide profiling of DNA-protein interactions by improving the traditional ChIP-Seq protocol and offering flexible barcoding options to facilitate multiplexing. ChIP-seq analysis is useful for identifying the cistrome of DNA-associated proteins and map global binding sites. The NEXTflex ChIP-Seq system facilitates this analysis by reducing the amount of time needed for library preparation and significantly reducing sequencing costs by allowing up to 96 samples to be run in a single flow cell lane.

According to Dr. Masoud Toloue, Director of Genomic Research at Bioo Scientific “NEXTflex ChIP-Seq covers a wide range of applications, from the measurement of transcription factor binding as a response to environmental stimulus, to clinical applications include monitoring embryonic stem cell development and cancer progression, yielding a functional annotation of our genomes. What was once a specialized assay, (ChIP) now can be applied on a genomic scale. To achieve this, polymerases and ligases used in traditional library preparations must be optimized to bind to minimal amounts of genomic input. The NEXTflex ChIP-Seq Kit utilizes NanoQ™ technology to achieve this.”

The enzymatic steps in the ChIP-Seq kit take advantage of Bioo Scientific’s proprietary NanoQ™ low input binding enzymes. NanoQ™ enzymes have been designed for use with DNA input material of 10 ng or less and may be applied to nano-sized microfluidic library preparations. The NEXTflex ChIP-Seq Kit also features “Enhanced Adapter Ligation Technology” which results in library preps with a larger number of unique sequencing reads by making improvements to the ligation enzymatic mix, the user will now have the ability to perform ligations with longer adapters and expect to see better binding efficiencies. The NEXTflex ChIP-Seq Kit simplifies workflow by using master mixed reagents and magnetic bead based cleanup, reducing pipetting and eliminating time consuming steps in library preparation. In addition, the availability of up to 96 unique adapter barcodes makes this the most high-throughput kit available. This kit can be used to prepare DNA libraries for sequencing using Illumina® GAIIx, HiSeq and MiSeq platforms.

    Strategies for Sequencing Challenging Samples
    by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
