    Using SOPAdenovo: no [LIB] in file

    Hello everyone,

    This is my first time to do de novo assembly. I'm trying to use SOAPdenovo-63mer to assembly a file.fastq from NCBI.
    However when I type this command:
    ./SOAPdenovo-63mer all -s /home/NGSusers/b99b02008/iGEM/SRR037531.fasta -o outputgraph -N 12000000 -V

    It told me "Config file error: no [LIB] in file". What is it mean???

    plz give me a hand. Thank you very much!!

    Hi Liyiyuian,

    It looks as though you are giving a .fasta file rather than a configuration file in the "-s" argument.

      Hi there!

      You need to create a .config file.
      ( http://soap.genomics.org.cn/soapdenovo.html Appandix A as an example)

        Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

        I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops.
