    Loading more than 10 pmol onto a two lane flowcell

    Has anyone tried running more than 10 pmol (the suggested amount) on the two land flowcell?

    We ran one run at 10 pmol and hit 250 million paired end reads. Even though that's right in the correct range we (my PI) wants to try and bump up our read count if possible. Has anyone played with this?

    Generally speaking (in my limited experience and assuming similarities with Miseq loading), the number of Q30 reads flatten out before the total reads i.e. you may load more and get more reads, but a lower fraction of the reads are actually useful.

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
