  • Ion Proton and fungal ITS amplicon sequencing.

    We have an Ion Proton that I will like to carry out fungal ITS amplicon sequencing on. I intend to use ITS1f and ITS 2 primers. However, while I have successfully carried out WGS on the Proton, this is the first time I will be doing amplicon seq. I do know I need to put in barcodes (as I have about 8 samples) and adapter. Would some one have a work flow I can modify or where I can get the necessary information? Is it possible to have custom made oligos with the adapter and barcodes? If so, how do you select your barcodes, their location on the oligos e.t.c?

    Ion has a protocol for amplicon sequencing on their Community site. Look for Fusion Library Protocol.
