  • preparation kit for low input poly-a selection RNA

    Hey guys,

    I want to perform RNA-seq on FACS sorted material. That means I will have some degradation and low input (around 10 ng and potentially less). Also I want to have quantitative data for differential expression analysis why I think poly-A selection is the method of choice. Does anybody have experience or literature on which preparations kits work best? Or a strong opinion about why total RNA might also work well using some particular kit?
    (We are talking about Drosophila here).

    The Clontech SMARTer v4 ultra low input kit works very well for total RNA input amounts as low as 100 pg in my hands. We have used it on material with lower RIN scores (6 and even below) with success. I would recommend this workflow if your goal is to interrogate poly-A transcripts. Once you make cDNA with this kit, you can go into the Nextera XT workflow as Clontech recommends or Rubicon's ThruPlex DNA Seq kit for library prep. Best of luck!
