  • Selection of indexes

    Hi,
How to select an optimal set of indexes from different library prep kits to design a sequence run?
An example: I am about to prepare libraries and sequence 40 samples prepared with XT dual indexing and 21 libraries prepared with single indexes (ScriptSeq).
Given a set of 8 i7 indexes from the XT libraries the task is to select a set 21 index among 48 available that will differ on at least two positions relative to the 8 i7 indexes.
It is quite tedious and error-prone to do this manually.
Does anyone have a piece of script to do this?
Or how do you select an optimal set of indexes?
    Cheers,
    /Jakob

    Hi again,
I have made a small R script (attached, "compare_index_Rscript.txt") to compare two lists of indexes (equal index length) and return the number of different positions per combination - thereby making it possible to select the most diverse sets.
    Cheers,
    /Jakob

      Current Approaches to Protein Sequencing
      by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
