I have a question about ssu rRNA gene fragment analysis when the amplicons are sequenced on NGS. In old times when we had 454 data, a singleton was considered to be 1 unique sequence. When I do now Illumine PE sequencing, I have the lowest number of reads 3, non of 2 or 1 unique sequences. Why it is so?
