hi!
how is it assured, that only one fragment per bead is contained in an emulsion-droplet? why canīt there be more different(!) template-fragemts per bead, which would lead to nonsense-sequences?

cDNA normalization

hi!
is it possible to do a cDNA-normalization prior to the sequencing with solid? the solid-protocol does not mention normalization, but i think it would be very useful for me. iīm not sure if the...

@kmcarr: we already performed a microarray - sequencing is the next step.
@all: thanks for your advises. seems that solid would be a good choise, but of course there remains the problem that there...

thanks krobison for your post!
our aim is to compare 10 different transcriptomes. these transcriptomes are derived from 5 different near isogenic wheat-lines, all fusarium-inoculated and...

its about the involved genes in fusarium-resistance. so i think longer fragments are never bad but i donīt know if we really need them in that case. annotating genes should also work when assembling...

wheat transcriptomes

hi!
we want to analyze 10 different wheat transcriptomes. we planned to use 454-sequencing, but iīm not sure if this is the methode of choice (because of the transkriptom-size, needed coverage and...

1/2 read and coverage?

hi!
sorry in advance for this stupid question, but i couldnīt figure it out anywhere:
what is 1/2 (or 1/8) run? does this only mean that half of the standard-time is used for sequencing, so...
