Dear all,
Has anyone tried re-washing prepared Ion PI chips after prepping and storing them for >24-48 hrs? Our FAS told us that it's ok to re-wash once after such storage and we have used chips...

NextSeq - ribosome profiling weird results

Hi all,

We received a set of ribosome profiling libraries to be sequenced on the NextSeq 500. We ran the pooled libraries using 1 x 40bp chemistry on the NextSeq. The pool was qPCR quantified and...

High polyclonality from Ion Chef

Hi,
Has anyone observed very high polyclonality from Ion Proton after the Qubit assay being in the optimal range for templated ISPs? We have pools of amplicon libraries that have some complementary...

Thanks for the reply! We actually ran two other samples in this same chip and those were not contaminated with this same peak. It's unlikely that only this pin was contaminated. We can try to redo...

Amplicon resolving differently between bioanalyzer and tapestation

Hello,
We are seeing a discrepancy between the Bioanalyzer and Tapestation for a large amplicon product that we received for sequencing. We received the PCR reaction for this amplicon and did an...

The hump after 400bp could be a bubble product...

The hump after 400bp could be a bubble product but the sharp peak at 250bp could be a population of RNA around ~125bp that is inherent to this sample or a contaminant of some sort. Remember that...

rRNA-depleted RNA profile looks weird

We received these bioanalyzer profiles after rRNA depletion (using Illumina stranded Total RNA kit, we check for removal of rRNA even though it's not in the protocol) from Th17 cell total RNA. The...
