Storage of Libraries

Just wondering,

Has anyone looked at the storage of their libraries? Are you storing at -20 or -80C? For how long? Have you pulled out an old library and progressed to a successful sequencing...

Paired End Sequencing on Ion PGM

I was wondering if anyone has ran a paired end run on the Ion PGM.
If so, what are you feelings about it? Is is a lot more work for not
that much gain, or it is worth looking into?

Thanks,

Right now, I am having to use the Roche Light cycler Z-480. If you have to buy an instrument, I would say go with one of Life Tech's. Maybe the 7500?

Good luck!

Hi JT,

I still would not trust the qubit numbers. The important thing here is "amplifiability", and the qubit does not tell you that. I always measure my FFPE DNA with the Qubit first, before I...

Hi Lindsay,

I see you are in Dallas? Where are you located?
I see you are putting 96 libraries onto one chip, but what are your libraries of?

I would say to keep with your qPCR. I have tried the equalizer, and it is just too much of a black box for me. Doing 96 samples may be a bear, but you can rely on the qPCR data. Can't really say...

You probably need to check amplifiability of your DNA. Using the RNaseP kit will help out with that. Crap in, crap out.

Hi JT,

I am pretty much doing the same thing as you, but I am getting very nice data. A couple of things to integrate:
1. RNase P kit to quantify your amplifiable DNA before PCR. This will...
