    Has anybody tried the Ovation Single Cell RNA-seq system?

    Hi,
    it would be interesting to hear how good it is...the kit costs about 5000 USD for 32 samples! In the FAQ sections they say the kit is designed for high-quality RNA (NOT whole cells, apparently...so why calling it "Single-Cell"?) and the amount of rRNA reads will be <25% (seriously? after paying all that money 1/4 of my library will be junk?).
    Besides, what are these nucleotide analogs that are incorporated during the first cDNA strand synthesis and later they are used for the fragmentation? Any idea? thanks!

    Bumping this thread because I am curious as well.


    We tried 500 pg of good quality total RNA with this kit. It had horrible results - 60-90% Duplication rate, 30% rRNA fractions. Anyone else have this experience?

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

      I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops.
