    Buy sheared DNA

    Hi all,

    I don't have Covaris, and need some sheared control DNA. Ideally, broad range, so I can size select. Anybody knows commercially available sheared DNA (available in EU)?

    You cam make your own fragmented DNA by using a cheap source of DNA and 6 or 4 cutter or combination of restriction enzymes. Other option is using NEB Fragmentase product: https://www.neb.com/products/m0348-n...na-fragmentase

    I do not think there is a high demand for fragmented DNA, so I never have heard of it. A commercial option is buying SageScience control DNA for Pippin systems. They are restriction fragments and you would need a clean up to remove their loading buffer before using for size selection experiment. http://www.sagescience.com/product-s...-prep-support/

      Many places sell sheared salmon sperm DNA as a blocking reagent. I'm not totally sure the size range is in the range you desire, but it might be!

        Thanks SNPsaurus for reminding Salmon DNA. The product size varies from vendor to vendor. ThermoFisher product size is ≤2,000 bp, Trevigen is 80-500 bp and Agilent one is 300-2000 bp. Other suppliers might have different size range.

          oh that' a good idea really! Thanks. I was thinking about fragmentase or some ZnCl2 digestion or so, but wanted to keep it simple.

              Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
