    How much of PacBio ZMW Loading Productivity 1(p1) is best for IsoSeq?

    Hi,
    Now I am investigateing the IsoSeq in PacBio platform. I find that the ZMW Loading Productivity 1(P1) varies a lot from different experiments. So I am confused what's the normal range for P1. And what's the effects for too low or too high P1? How does it come out for that?

    First, our recommendation for loading for different applications can change over time, so please always check with your local PacBio FAS or sales person to get the most up-to-date answers.

    Currently, for RS2 data, we recommend loading P1 between 50-70%. Some customers have had success loading above 70% but in-house we observe decline in base quality after 70%. Lower P1, by the way, does not necessarily improve base quality, but you get less yield and less bang for the buck, that's all

      Now we have a lab the loading p1 has been above 70%:
      Prod=0 ZMWs: 9.675%
      Prod=1 ZMWs: 76.32%
      Prod=2 ZMWs: 14.01%
      Total Active ZMWs: 90.32%
      I can not figure out it is good or not. Is there any way to evaluate the quality of the data?


      I have evaluated data quality using StsPlots Tool. But I don't understand what does the fiugur mean. So is there any one can help to tell is there any problem in my cell?
