    Maximum number of cycles for kit Miseq 50 cycles

    Hi sequencing Masters!

    What is the maximum number of cycles we can do with a Miseq V2 50 cycles kit. I have been told that there is enough reagents for more than 55 cycles including the 6 cycles for index reading. Can someone confirm this information?

        It should be fine. Local core does 66 cycle (50x8x8) runs off and on.

            I got an answer from Illumina's tech support:
            MiSeq v2 contains enough reagents for 75 cycles of chemistry

              I know this is late but we routinely do 78 cycles with the MiSeq v2 50-cycle kit. (31 read1, 31 read2, 8 index1, 8 index2).

                Plus 7 dark cycles used for index2 read so 85 cycles in total. This seems to be 10 cycles more than Illumina specification.

                  Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
