    Miseq- error

    Hello,
    My miseq showed an error and stoped running two hours after the run started.
    Error: Through- focus peak did not exceed SNR threshold (Illumina.Instrument.Bolt.Logic.AutoFocus.DarkSampleException)</ErrorDescription>

    It was using a V2X 500cycles kit with 24 samples from SOPHIA Genetics NGS Panel.

    Could you explain me which could be the reason and what could be the solution?

    Hi Maria,

    I'm not familiar with this exact error message but it sounds like the machine may have had difficulty finding a plane of focus for the flow cell.

    The very first thing you should do is contact Illumina tech support and they can help you troubleshoot your instrument and failed run. If you need additional help troubleshooting libraries, you can reply to this thread and I'll see if I can help you.

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
