    nextera xt for 30 Mb genome

    Hi all,

    I was wondering if anyone has experience or has tested using the nextera xt kit for moderately sized eukaryotic genomes (20 - 40 Mb).

    Any feedback would be greatly appreciated!

    1 ng DNA of the organism will have 23,200-46,300 genome equivalents. Assuming 1Kb average fragment size and 25% conversion rate 1 ng DNA will result in 231M unique fragments. If you sequence the library below the unique fragment numbers to obtain the intended coverage then it should be fine.
