    MiSeq Error in LanePump

    Hi All,

    We just ran a MiSeq 2x300bp amplicon sequencing run over the weekend and it has completed with decent specs but has shown an error that says the following:
    "Error in LanePump. Syringe pump value failed to move. Valve overload. Command = '/1Qe\r', reply = 'j'.

    I've seen most other MiSeq errors before, and have already sent an email to illumina tech support. I was wondering if anyone else had experienced this before -if you have any insight into how it may have impacted the output etc.

    Update:
    Tech support have identified it as a software or hardware error and told me to power cycle.
    Unfortunately its a hardware error so now I have an engineer coming to fix the machine.

      Thanks ikripp,
      I got the exact problem few days ago and already called tech support. Just wonder to know how did they fix it - replacing the pump?

        They ended up replacing the hard drive and that resolved the issue. I believe the pump error was a result of the hard drive failing. Tech support instructed me to power cycle the machine and when I did it wouldn't turn back on! Then they came and replaced the hard drive and we had to reconfigure the machine again.
        On a positive note it is now running smoothly!

          Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
