    Hello and dark digit cycle on HiSeq and NovaSeq

    Hi at all,

    im new in the forum and i want say just hi. Hope we have a nice time.

    Anyway, my question is about the dark digit cycles on HiSeq and NovaSeq.

    First, what does it mean and what is happening in this cycles??
    How many cycles are dark digits and so on...

    Look on page 5 of this document from Illumina. I believe you are referring to "chemistry-only" cycles described there.

        Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
