    16S & 18S Deep Sequencing Request(s)

    Hello,

    We've done 16S/18S/ITS/amplicon sequencing for years, with good results - sometimes excellent (library dependent) - for both internal and external clients.

    We're starting to see more and more requests for guaranteed extremely high read depth per sample - like in the neighbourhood of 50,000 to 100,000 reads per side per PE sample. What kind of Illumina instrument (other than a MiSeq) does this (amplicon) sequencing well and why are people becoming obsessed with ultra-high read depth?

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

      I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops.
