    Homemade illumina oligos and library prep kit

    Hi,

    I am wondering if we can use the PCR amplification primers which were made at our side.
    In theory, this can work properly.

    But I am worrying a little bit if there can be some problem with sequencing primer binding and PCR amplification step.

    Is there anyone who attempt this?

    Yes, as long as the sequences you use are the same as the Illumina sequences, primers you synthesize yourself work fine!

      Is this also true for adaptors and sequencing primers?

        For the PE PCR primers, is it necessary to PAGE purify them?

          Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
