    Paired End Protocol for Very Low Abundance Samples

    All,

    This is my first post after sometime lurking. Utilizing what I've seen in the forum I think I have suitably adapted the standard Illumina ChIP paired end protocol to achieve our goal of generating a sequencing library from 0.3 -0.5 ng input. The changes are not surprising (reduced vol. ligation step, diluting adapters, 20 cycles PCR enrichment, etc).

    Has anyone come across a reliable protocol or perhaps have any other suggests. Thanks everyone.

    I tried to respond directly, but your were not accepting emails. Yes the final conc of PE adapters I used for 1ng was 1.67nM.

      would either of you please share your protocol with me? I have been thinking of ways to make PE libraries from low input DNA (in ng). I will really appreciate it.
