    Alternatives to Roche Shotgun and PE protocol

    Hi,
    I hope you can help or give some pointers in the right direction. we've been working with Junior recently and have tried genome sequencing using the 8 kb pair end, it worked, which was nice but it was a lot of work and took a long time.

    I was wondering if anybody has come across alternative protocols that may simplify or speed up either the pair end protocol or the shot gun protocol.

    I have come across the cambio enzyme chemistry alternative and the beckman SPRI works robot for 454, which both look promising for the shot gun alternative?!

    Any other suggestions are more than welcome!!!!

    Roche's "paired end" libraries are what would be called "mate pair" on the Illumina or SOLiD platforms. These are quite challenging libraries to construct.

    However the shotgun libraries are not too bad. Are you using the "Rapid" protocol?

    Epicentre has a kit that might be what you are looking for:
