    Experience of Illumina TruSeq Exome coverage?

    Hi,
    I'm wondering what coverage others are seeing with the Illumina TruSeq Exome, specifically % bases with either 10, 15 or 20x coverage. We're running 6 samples in a pool, across 2 lanes - any idea what we should expect? An average measure isn't as helpful, since I'm more keen to quantify what we're missing.

    Roughly 2/3rds are greater than 10X coverage when a single exome is run in a HiSeq lane. This wasn't a v3 run though.

      Truseq 62 Mb exome enrichment

      With the 62 Mb exome enrichment, on a hiseq lane we 6-plexed and got ~70%PCTbases > 20x

        Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

        I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops.
