    Comparison of 454, Illumina & Ion Torrent

    Hello,

    Does anybody have an actual overview of the pros and cons of these sequecing methods. I have to give a presentation on this issue, but they improve so quickly, that i cant find the latest comparsion of this 3 methods.

    Performance comparison of benchtop high-throughput sequencing platforms.

    There is a bunch of net stuff here and elsewhere on critiques of the paper (e.g. mine, which has some links to others). Perhaps the most serious issue is that the Ion protocols in particular are much newer than then kits used in the paper, so results should be better on Ion than presented -- but how much better in a typical lab's hands still remains to be established (and by then the kits will have changed again! )

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

      I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.
