  • DNA Shearing

    Hi everyone,

    I would like to fragmentate/shear the DNA samples into small fragments prior to the target capture of interested genes. Plan to use sonicator to do it. Do you guys have any idea how should I do it? Can it be done by using the normal probe sonicator in the lab? I think I need to optimize it to get the favourable fragment size.

    What make/model sonicator do you have? If you post that info here someone may be able to give you the conditions you need.

    We use the Covaris to fragment genomic DNA into ~200bp fragments. If you can get a consistent temperature in the water bath then it's very reliable in terms of the fragments sizes that are generated.

      the only sonicator we have here is sonicator utrasonic processor XL and it has never been sonicated DNA before that's why I'm not sure whether I can use it or not.

      Covaris is pretty good based on what I read from the literature and publications just that we don't have it here. Thanks anyway.

        I'd recommend the Covaris. Just keep an eye on the water levels and make sure the temperature is within guidelines

          sonicator

          A probe sonicator will work fine. You must match the probe diameter to your sample size. The XL is most likely #XL-2020 from Misonix. Contact me if you want help with probes. [email protected]

          Or you can go to sonicator.com and review the probes that are available. You can match up what you have to the charts to see if you already have the proper equipment.
