  • questions about Illumina mRNA sample prep

    Hi,
    We are currently using a standard Illumina mRNA protocol and have experienced two problems :

    i) some genomic contamination
    ii) inefficient depletion (using the Ribo- kit)

    Has somebody experienced the same problems and has some words of advice?

    To remove genomic DNA contamination you might try TURBO-DNA free from Ambion before Ribominus treatment. This approach has worked well for us.
    Unfortunately, the Ribominus kit does not remove all rRNAs so you'll still see some contamination, especially if you have a little degradation of your RNA
    preparation.
