  • Good alternatives to Illumina TruSeq DNA Nano HT Kit?

    Hi,

    I am currently considering switching from Illumina's TruSeq Nano DNA HT Kit to another DNA library prep kit (it has worked great for us, but has unfortunately become unaffordable). Has anyone made this switch successfully? The kits I am currently considering are Lucigen's NxSeq® AmpFREE Low DNA Library Kits and Adaptors, NEBNext UltraDNA kit and an in-house protocol.

    We will be using Illumina HiSeq2500 to sequence these, and *hoping* to pool more than 96 libraries (maybe increase to ~125 libraries on one lane).

    If anyone has had success with any of these kits or has some good recommendations, I'd love to hear any feedback/thoughts!

    NEBNext Ultra II DNA Library Prep kit is as good as TruSeq and produces good libraries down to 500 pg inputs. Its workflow is simpler than TruSeq with less prep and purification steps. However, I am not sure if more than 96 pooling is possible or not.

      Kapa Hyper, Qiagen QiaSeq, BiooScientific NextFlex Rapid kits, will all work great with adapters synthesized by third parties or the extensive NextFlex adapter offerings.
