Best tool to look for alterntive splicing

Dear all,

I would like to know which tools you use when you want to look for alternative splicing events.
Some goodies I am particularly looking for:
- works with either single or paired-end data
- possibility to discover new junctions (not only reference-based)
- possibility to quantify the events with pairing between samples (not only by group but also by treatments, sequencing dates, etc...)

I saw few but none has all of this (particularly the pairing analysis), so I am asking the community
