  • QuBit fluorimeter vs. Epoch?

    Dear all,

    I measured DNA concentration with two way, QuBit and Epoch.
    For NimbleGen exome capture in Illumina sequencing, DNA is needed 30ng/ul.

    But DNA concentration measured by Epoch (900ng/ul) was way higher than DNA measured by Qubit(700 ng/ul). I used DNA BR kit for QuBit.

    Is Qubit more reliable? I assumed that epoch cannot detect low concentration well.

    I am afraid none of them is reliable. When possible use the most conservative value.

      we use Qubit. We find it gives values slightly higher than the bioanalyser but we base our conc's and experiments on the Qubit readings and haven't had any problems. We also use Qubit and bioanalyser after fragmentation...basically whenever the protocols say bioanalyser we also do Qubit.

        For 30 ng/ul I would use the HS kit on the Qubit.
