  • Can a RIN be obtained after running mRNA- instead of total RNA assays on Bioanalyzer?

We ran mRNA instead of total RNA assays, and are left with rRNA contamination values instead of RINs. The traces of the RNAs all look fine, so it seems that the data are there, and all we need is to find a way to get the RIN from them. Is there a way to (non-manually) obtain RINs after running mRNA assays? I also posted this question as a response to a related thread in this forum, but decided to create a new thread in addition, because it seems like a problem that is not unlikely to occur to people doing library preps. Thanks for any help.

No, I don't think this is possible. But if you figure out a way, please post it.

I recall reading that RIN is the value assigned mainly based on rRNA peaks. So even if you'd get RIN value for your samples it might be wrong because without rRNA peaks the algorithm might misinterpret the RNA integrity. This is how I understood it.

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
