  • Barcodes and adaptor

    hi there,

    I have two questions:

    1-What is the mechansim with which Ion barcodes and P1 adaptor works?

    2- why there is only one adaptor instate of two?

    Hi there,

    There are, in fact, two library adaptors - we call them A and P1. In most library preparations these are ligated onto library insert fragments. In some cases, these may be amplified onto target sequences via PCR. If you currently are using the PGM or Proton, your local Field Applications Specialist can readily supply more information. I would also recommend consulting the Ion Community forums as a number of threads talk about adaptor composition.

      I personally find the Ion Community's website annoying due to it's obsession with controlling access. Can someone post the adapter information here?

        Hi there,

        Ion Community is completely open to all registrants - i.e. there are no member access levels. You should have full access to all technical information contained within.

        With the present revision of the fragment library user guide, the adaptor structure may be found on pp. 70:

          In my lab they use two barcodes but the adaptor is (P1) and there is no (A) adaptor. I do not understand why???? I would like to know if some one uses this

            the manuel you put a link to uses 13 pb adaptor barcode, which is a new one I am not sure that we use in the lab.
