  • Streptavidin Bead Quantitation

    Hi Everyone!

    I am trying to evaluate how well our new robot handles bead clean-ups with streptavidin beads. I would like to objectively quantitate bead carry over (after removal of supernatant) rather than just visually assessing. Any ideas?

    Counting with a hemocytometer.

      more context is needed, but you could:

      1. use A600 as a measure of particulate carryover within the supernatant
      2. pre-bind beads with biotin-fluorescein, then use biotin to displace to get carryover in terms of biotin-binding equivalents using a standard curve (fluorescence reader required)
