  • Ion PI™ Controls usage with Ion Torrent PGM

    Hello,
Does anybody know about compatibility between control sphere from Ion PI™ Controls 200 Kit and PGM in terms of sphere size, sequencing primer and so on? I need a control sphere for PGM but it looks like there is just Ion PI control kit with premade libraries and spheres. Is there any other control sphere kit for PGM?

    Hi,
you're right, the v3 version of the Ion PGM™ Controls Kit doesn't have any Control Ion Spheres. The v2 version did.
Well size is notably bigger in PGM chips compared to Proton's PI, so I would say that the ISPs are not interchangeable, unfortunately. You might end up losing them during the run, I suppose.
An aliquot of Control ISPs comes with PGM sequencing kits - although they are barely more than what's needed for the kit's normal usage.
Did you need to make a test run with only control ISPs, as opposed to the normal amount of control ISPs that go in a sequencing run? Thermo engineers do use full control runs to check the machines at installation, so perhaps Thermo might be able to help?

Thanks a lot. Yes I need to run full chip with control IPS.

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
