I have to do a WGA from few cells (10 or less) and I need to strip off all the proteins from the DNA before starting with the protocol. I came across a kit from ZyGEM which contains EA1 proteinase (+ Ak1 also, I think). This proteinase has the advantage that it gets completely inactivated by a short inactivation at 95 deg. (contrary to proteinase K which needs harsher conditions in terms of salt and denaturing agents), it works in common buffers and doesn´t need to be removed before proceeding with the following enzymatic reactions.
I think that EA1 is the same contained in some (maybe all!) WGA kits, such as the Picoplex from Rubicon Genomics.
My question is: does anybody know if any other company is selling the EA1 proteinase separately or do I really need to buy the kit?
