  • Miseq V2 kit (2x250 cycles) drop quality

    Hi,

We detected some drop quality (R2) on the Miseq V2 kit (2x250 cycles), anyone else?
    p. ex. R1 finished arround 87% Q30 and R2 with 67%.

Is the V2 kit the new (old) V3 (2x300 cycles)?

    Cheers,

For all discussions about MiSeq quality it is essential to include information about the type of library, cluster density for the run, spike-in type and concentration used :-)

      You're completely right

      Lib prep : Nextera XT
      Samples: Mixed genomes no Phix

You chances of getting a free replacement kit from Illumina would be slim for this one The drop does not look too bad. Could be library pool dependent in this case. Perhaps it is only some of the samples from the pool that are dragging that total number down for R2. Have you checked?

The replacement for this kit arrived today
We'll receive another kit, from other Run, no more replacements because of the high cluster density

This kit its very stable even in overclustering mode, but now looks like the old V3.

What does quality by cycle look like? No PhiX makes it hard to nail down if it's an issue with your pool

              Current Approaches to Protein Sequencing
              by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

              Strategies for Sequencing Challenging Samples
              by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
