  • What is the meaning of repeat cluster and unique cluster?

    Hi

I am very beginner in Bioinformatics especially sequencing method.

Now I just read some paper (Title : De novo assembly of human genomes with massively parallel short read sequencing)

So I can't understand several terms in the paper. So I need your help.

    In the paper contents,

"Resolving the repeat clusters between unique clusters and assembling them
into an intact sequence requires the paired-end relationship of a pair of short
reads generated from both ends of a DNA clone"

In this paragraph, I can't figure out that the term repeat clusters and unique clusters is indicating about. I have learned that cluster is just a action that gathers something similar. But in this paper, what is the repeat clusters and unique cluster meaning?

And also, I am not sure of meaning of paired-end method.

Since I am very beginner in this areas, so I am looking forward to hearing some easily explain about my questions.

    Thank you

    Strategies for Sequencing Challenging Samples
    by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
