  • Throughput of Truseq SBS v2 kit??

I could find v1 and v3 but not v2. Anyone knows?

    Thanks in advance!

Found this Illumina ppt claiming it goes from 250Gbp to 600Gbp:

    Perhaps, v2 looks like this?

    HiSeq 2000 v2 dual flowcell 8-lane/flowcell

    1x35 1.5days 33-44Gbp 940M-1.25B 90%Q30

I don't remember there being a v2. Chemistry seemed to jump from v1 to v3 for highoutput HiSeq (8-lane flow cell) chemistry.

Another piece of evidence that supports the existence of v2

It's been long enough that I've forgotten most of the pre-v3 information that I used to know, and the notebooks are all in storage at this point.

          I did find this:

which reminded me: the cluster generation kits went through a couple of iterations between SBS sequencing v1 and v3. IIRC, there was such a thing as a v2 cluster kit, that worked with the v1 sequencing reagents. At the time v3 SBS reagents came out for the HiSeq, the cluster kits went to v3 since the increase in sequencing output was at least partly due to some flow cell changes.

Another paper used the SBS v2 kit:

Yes, given the evidence you provide, I guess there was HiSeq v2 SBS chemistry. The only time we used v1 chemistry was during the install run on our instrument. For an install run, Illumina must have just been using up there stores of v1 chemistry at that time.

                Strategies for Sequencing Challenging Samples
                by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
