Default how do you find a possible intron on a gene?

hello all, i am relatively new here. I am wondering how do you guys find a possilbe intron on a gene? what are the criterias? what software do you use?

What do you have? Do you have the sequence to an unassembled genome? An assembled genome? The sequence to an RNA molecule?
