  • Hiseq 1500 run quality issue

    Hi,

    We have got a Hiseq 1500 instrument over 3-years and we are happy about the throughput and performance. We have run the instrument quite frequently at least one run per month. However, the recent run performance is not stable. Q30 of read 1 of all lanes were fine but index read of two lanes were abnormal. While Q30 of read 2 were various and they were not good in general.

    We have checked the volume of water wash and maintenance wash (before and after the run) was normal and even. It indicated the fluidic shouldn't be the problem.

    We have contacted Illumina for help and they recommended us to use NaOH wash to resolve the problem but we concerned it wasn't total solution to the problem. Actually we have done NaOH wash before the run with problem.

    The cluster density is around 994 to 1050k/mm2, the run mode was high output run PE100. Most of the libraries were RNAseq and we have run the similar libraries successfully before.

    We wondered is there any similar experience among other users who have got Hiseq 1500?

    Is it the aging problem of the instrument?
