  • Solicitations Regarding the Recent Upgrades to 120 tile format

    Hello.

I am trying to get a sampling of peoples experiences concerning the recent "upgrades" on the GAII machines that incorporate the new reagent/cooling system. We have several machines which used to function marvelously. Since Illumina started sending people out to "upgrade" them they have never quite worked right. We see much worse map'ability on 2 x 76 human RNA-Seq libraries suggesting base calling issues, and the signals are dropping dramatically during the course of the runs. (They are not overloaded, 12 million clusters and v 1.4 software). In short, we got much better results when we had to use fresh reagents that we changed more frequently.

We are inclined to think it is either mechanical or chemical in nature. I'm wondering if it has to do with the new cooling/reagent system that has recently been installed?

What have other people's experiences with the upgrades been? Love 'em hate 'em?

    Hello,

You post caught our interest as we are about to upgrade our GAII machine. It's rather new as its only been functional for a few weeks. Can you share more details with us? For example, when you say:
the signals are dropping dramatically during the course of the runs
how many bases can you effectively use? 2x50?

    Thank you!

      Strategies for Sequencing Challenging Samples
      by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
