Hi,
We are successfully using a Covaris sonicator to shear DNA for ChIP-Seq on Solexa platform. Especially the reproducibility is higher than that of other systems we have tested so far. Only the cost price is a major drawback. Best andibody |

Well, buy a SOLiD then, and you will get a Covaris also.

I did see they now have a 96well version as well. I'm slightly worried that the biorupters sonication heats the sample and we could get biased shearing. On the Covaris website they state that this method is isothermal. Did anyone check this yet?

does anybody have experience with the KASPar genotying assay from Kbioscience? http://www.kbioscience.co.uk/reagents/KASP/KASP.html I am thinking of using this technology but I first would like to know your experience. Thanks a lot! |

I'm looking to create a couple of 8kb paired end rapid libraries for 454 seq and don't have a HydroShear, I realize there are a few places to outsource the shearing step to facilities such as Profile Genomics or Covaris but has anyone tried Covaris's g-TUBE single-use device that shears genomic DNA into selected fragments sizes ranging from 6kb to 20kb. The only equipment needed is a compatible bench-top centrifuge, looks like it would be about $27/sample.
