  • All Data Disappears After Run

We had our 454 upgraded several weeks ago. Now when the run completes something deletes all of the data files from the instrument. The log files show the run was completed and the data and image files were written to the disk, but they are just not there. This has happened twice since the upgrade. Has anyone else had their data disappear like this or have any idea what could be happening?

You or your technician did not install/edit the backup script which is ran at the end of sequencing. It is usually used to copy data from the sequencing machine (it's computer) onto a server machine. Look into /data directory on the sequencer machine.

Well, this is usually the 'postAnalysisScript.sh'. This should be edited to match your needs/environment. As martin2 has already mentioned, have a look at /data on your machine. AFAIK the system (default) is not configured to automatically remove runs from the systems when they have been (successfully) copied to the analysis server.

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
