  • Problem in primary analysis

    Hi, I have one problem I can't solve myself.

    I was making run on Solid, and for some reasons solid cluster was rebooted.
    So, instrument was stopped and then resumed. After resuming, imaging and fluidics was OK, but there were no bindFinding and color calling - so now I have only focal maps and images, but don't have any intermediate results such as satay reports and color calling.

    How can I restart primary analisys from raw images? SETS says that primary re-analysis can be made only from color calling, not images.

    So, I think it's the same question as "How is the image processing started directly on SOLiD?"

    I will be very pleased if you help me

    With issues of this sort we contact Applied Biosystems. They can login remotely, if you allow it, and start image analysis for you.

    If this is not possible, I have a way that might work: re-do the focal map. Does not cost much (you can steal the reagent from a WFA kit.) This does not just complete image analysis, it would repeat it from the beginning. We did is once and it took >3 days (~ 1 hour/cycle reprocessed) to completely re-do image analysis! More detail in an earlier thread:



    But getting AB to re-start image analysis for you, would be a much better method with less risk.

      Thanks a lot!

      I have written to the AB specialists to solve this problem, but maybe it's too early now in their time zone to answer me.
      Nevertheless, I'm unpleasantly surprised that fact that there no convenient method to restart imaging locally. Maybe it's because of very little probability of this issue...

        Yes, it is rare. Also, because doing image processing is computationally intensive, they may prefer to make it difficult to do -- so it does not happen unintentionally. Or due to a misunderstanding.

        Don't you have tech support you can phone?

          Yes, I called them (to our, russian representatives of ABI), but they also don't know how to do it. I think, they are going to contact with their colleagues. So, I hope it's a matter of time...
