  • Quality Control Library using QUBIT... help me!

    Hi all,

    I did the quality control of library using Qubit and FAM/Cy5 probes.
    Can someone tell me how ocurrs that?

    Where FAM hybridizes? Where Cy5?
    What is the ratio calculated?

    Hi there,

    This assay estimates the ratio of templated (positive) to non-templated (negative) Ion Sphere Particles (ISPs) via fluorometry with the Qubit. Non-templated ISPs are measured via hybridization of a FAM-labeled oligo to the primers on the ISPs. Templated ISPs are measured with a CY5-labelled oligo to the A adaptor end of clonally amplified templates. A simple spreadsheet is employed to perform the calculations and guide next steps.
