We have generated long amplicon library that is about 1400bp. And their diversity is high. Does anyone have experiences of sequencing such long amplicon on HiSeq4000? Any suggestions are welcomed. Thank you!

Is this a one-off experiment or are you planning for multiple samples?
You will need to shear the DNA sample either by sonication or enzymatically to pieces of about 300 to 450 bp length for the Hiseq 4000. Then it can be treated like a genomic DNA sample. |
