Does anyone know whether the Small RNA Sample Prep v1.5 kit can be used to clone and sequence longer small RNAs (say 40~50nt long small RNAs)? From Illunima website, it seems Genome Analyzer II can generate reads longer than 50 bases for genomic DNA sequencing. Is this possible for small RNA libraries?
Thanks in advance! |

yes the kit will allow you to capture longer fragment, but you are likely to get a lot of tRNA and not microRNA

Degrade uncapped RNAs and then decap or you will sequence tRNAs.

just wondering, but has anyone of you been able to capture miRNA hairpins or other RNA hairpins from an RNA mixture? I have been looking for a method to do this, but so far have not found one...
