  • Alternative sequence enrichment methods

I am looking into alternatives to NimbleGen sequence capture prior to Roche/454 GS FLX sequencing. Does anybody have experience of using the following sequence enrichment methods? Can they be used successfully for Roche/454 GS FLX sequencing?

    Raindance

    Agilent SureSelect (possibly Illumina only)

    Any other alternatives?

We are going to test Combimatrix's arrays in the following weeks.
Have you ever tryed any of the methods you mentioned before? How did they work?

I checked Combimatrix website, but am not sure how their exon capture works. Could you share some detail?

They do not have a sequence enrichment product (array) out yet. What we are trying to do is testing their arrays to see if they could be usable (and most interestingly for $ , REUSABLE) for seq enrichment. The method of hibridization would be the same as with Nimblegen's or Agilent's arrays.
Unfortunately the samples are not back yet.. so I ll keep you updated when we have some results from the sequencer

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

          Strategies for Sequencing Challenging Samples
          by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
