Smile About identify the diffenential expression gene

Hi,everyone
Currently ,We use SOLID to study transcriptome . I want to ask how to identify the diffenential expression gene ,just use the result of RPKM or some models ? Sorry , I am a layman on the bioinformatics .\

We also have some videos that go through the analysis of expression data, step by step. You can find these at - http://clcbio.blip.tv/
One of them is even in Chinese ;-)
