  • mRNA seq to multiplexing

    Hello All:

    Has anyone started with the mRNA seq protocol and then took it through the multiplexing protocol at the "Perform End Repair" step? Please share your ideas on how it was done.

    We would like to be able to do multiplexing in our lab, starting with RNA samples...

    Hi,

    We are going to sequence RNA-Seq libraries with indexing by preparing individual RNA-Seq libraries using the "mRNA-Seq 8 Sample Prep Kit (cat# RS-100-0801) followed by "index-PCR" using the primers of of the "Multiplexing Sample Preparation Oligonucleotide Kit (cat# PE-400-1001).
    At the step of ligating the PE adapter to the 'A' tailed DNA fragments, use 1ul of the Index PE Adapter Oligo Mix instead of 1ul of the PE Adapter Oligo Mix. Then proceed with the multiplexing cluster generation & sequencing protocols.
    And use the multiplexing sequencing kit when sequencing your libraries on the GA....
