Can anyone fill me in on the most cost-effective strategy from going from cell lysates to mRNA, either directly or indirectly? Currently, my lab is:
1) taking cell lysates and isolating total RNA (Qiagen RNeasy mini kit with Qia on-column DNase treatment)
2) running a gel to visualize RNA integrity
3) quantifying total RNA (Qubit) for steps 4 and 5
4) ERCC spike-in
5) Dynabead mRNA purification
With the RNeasy kit and the Dynabeads, there's a lot of money going in, especially with the RNeasy columns, which are not reusable.
Do you have any recommendations for alternative strategies?
